Hi!
Posting this to help others. System: FF 10.0.2, Win 7x64.
Had the problem that left-clicking on internet links wouldn't work anymore (to be more exact: you have to click a link 10 to 15 times to get through) unless I deactivated NoScript (2.3.4; retried with 2.3.4rc1, same problem). Tried to eliminate possibilities by successively switching off and on addons, found out: it's NoScript and TryAgain (3.4.6) that are biting each other.
Switch off both or either one - no problem. Run both at the same time: problem comes back.
Lost an hour off work with all this bs. Hope I can spare that other people!
I already informed the guy behind TryAgain by Mail - but some people might also be looking here.

Just tried, and it works for me on a clean profile with just those two extensions installed.
Does it for you?
Any further configuration detail you can share?
Does NoScript Options|Export + NoScript Options|Reset help?

Sorry, Giorgio, I've got a lot of pressure right now, and time's precious. But I've been using NoScript for some time, so OK - after all I'm indebted to you 
Tell me which details you need. Best if it can be done with an automatically created log. If it contains a lot of details, however, I'd rather not post it publicly, but send it to you by PM.
And yeah: I exported my whitelist and reset the options - no change. The only thing that helps is switching off either NoScript, or TryAgain, or both.

Could you also check whether any message appear in your Error Console (Ctrl+Shift+J) whenever your left click fails?

Works fine using the latest build and matching the specs you mentioned in your original post, so there has to be something on your end, try a clean profile and see if the issue persists. Then try standard diagnostics and see what works or breaks it. Cannot reproduce your issue.
